Early results from the phase 1 META 10-19 trial presented at the 2026 ASCO Annual Meeting showed encouraging outcomes for patients with high-risk relapsed or refractory diffuse large B-cell lymphoma (DLBCL). Among 14 evaluable patients treated with ultra-low doses of the IL-10–expressing anti-CD19 CAR T-cell therapy, the objective response rate was 100%, and 92.9% achieved a complete response.
After a median follow-up of 27.4 months, one-year progression-free survival was 71.4%, while one-year overall survival reached 92.9%. Several patients maintained their responses for more than 30 months, including those with central nervous system involvement, suggesting the therapy may provide durable disease control in a high-risk population.
The treatment showed a manageable safety profile. Although all patients experienced cytokine release syndrome, severe cases occurred in only 7.1%. Severe neurotoxicity was reported in 14.3% of patients, and no severe cases of IEC-HS were observed. Researchers believe IL-10 expression may improve CAR T-cell activity at ultra-low doses, supporting further clinical development of META 10-19.
