Anito-cel CAR T Therapy Shows 97% Response Rate in Refractory Multiple Myeloma

Anito-cel is a novel CAR T-cell therapy for multiple myeloma that reprograms a patient’s T-cells to target BCMA (B-cell maturation antigen). It uses a fully humanized D-domain binder for enhanced uptake, faster release from targets, stronger CAR expression, and lower risk of immune-related complications.

In the phase 2 iMMagine-1 trial with 117 heavily pretreated patients, anito-cel achieved a 97% overall response rate, with 68% achieving complete or stringent complete responses. Among evaluable patients, 93% reached minimal residual disease negativity. At 12 months, estimated progression-free survival was 79.3% and overall survival 95.2%.

The safety profile was manageable, with mostly mild cytokine release syndrome (Grade 1) occurring around four days post-infusion. Severe neurological events were rare and resolved over time.

With strong efficacy, a favorable safety profile, and improved access to CAR T-cell therapies, anito-cel represents a promising new option for patients with difficult-to-treat multiple myeloma.
