Abemaciclib and Letrozole Combo Shows Promise in Advanced Endometrial Cancer

Findings show that combining abemaciclib (Verzenio) with letrozole is an effective treatment for advanced or recurrent endometrial cancer.

The study found strong disease control, especially in patients who had not previously received chemotherapy. Overall, 56.9% of patients were progression-free at 6 months, rising to 71.4% in chemo-naïve patients. The median progression-free survival was 9.5 months overall and 12.7 months in patients without prior chemotherapy. The treatment also showed a 39.2% response rate, including some complete tumor responses.

The combination works by pairing hormonal therapy with abemaciclib, a CDK4/6 inhibitor that helps overcome resistance and improves the durability of treatment effects. Side effects were consistent with known profiles, mainly fatigue, diarrhea, and low blood counts, with no new safety concerns reported.

Overall, the results suggest this drug combination may offer a chemotherapy-free option for some patients with endometrial cancer, providing meaningful and longer-lasting disease control.
