10-Year Study Confirms Chemo-Free Option for Follicular Lymphoma

The final 10-year results from the phase 3 RELEVANCE trial show that the chemotherapy-free combination of lenalidomide plus rituximab (R²) provides long-term outcomes similar to standard immunochemotherapy for patients with previously untreated advanced follicular lymphoma. The study included 1,030 adults and followed them for a median of nearly 10 years.

Patients treated with R² had a median progression-free survival of 110.6 months, compared with 102.8 months for standard immunochemotherapy. The 10-year progression-free survival rates were nearly identical at 46.4% and 46.6%, while overall survival at 10 years was 82.4% with R² and 81.1% with immunochemotherapy. Time until the next lymphoma treatment was also similar between the two groups.

No new safety concerns were identified during the long-term follow-up. Rates of second primary cancers, disease transformation, and deaths were comparable in both treatment groups. These findings confirm that R² is an effective long-term, chemotherapy-free first-line treatment option for patients with advanced follicular lymphoma.
