P-32 microparticles (OncoSil), an implanted radiation device, combined with FOLFIRINOX chemotherapy showed encouraging results in patients with unresectable, locally advanced pancreatic cancer in the phase 2 TRIPP-FFX trial presented at the 2026 ESMO Gastrointestinal Cancers Congress.
The study enrolled 88 previously untreated patients who received either FOLFIRINOX alone or FOLFIRINOX plus P-32 microparticles. Both groups achieved similar 16-week local disease control rates, meeting the trial’s primary goals.
Patients receiving the combination treatment showed higher response rates (55.6% vs. 37.2%), longer median overall survival (18.2 vs. 15.8 months), longer progression-free survival (12.1 vs. 9.9 months), and greater tumor shrinkage. Surgery rates were similar, but the combination group had a higher rate of complete (R0) tumor removal.
The treatment was generally well tolerated. Serious side effects were slightly more common with the combination therapy, and 14.6% of patients experienced severe events related to the P-32 device or its implantation procedure.
