The phase 3 frontMIND trial has demonstrated that adding tafasitamab (Monjuvi) and lenalidomide (Revlimid) to standard R-CHOP chemotherapy improves outcomes for patients with newly diagnosed, high-risk diffuse large B-cell lymphoma (DLBCL).
After a median follow-up of 35.2 months, the combination reduced the risk of disease progression or death by 25% compared with R-CHOP alone. Two-year progression-free survival was 71.1% versus 62.9%, while three-year event-free survival reached 61.2% compared with 54.8% for standard treatment. Interim overall survival data also favored the experimental regimen, with three-year survival rates of 81.1% versus 77.8%.
Although severe blood-related side effects were more common with the combination, treatment discontinuation rates due to adverse events were similar between groups. Researchers say the results could expand first-line treatment options for high-risk DLBCL.
