The global phase 3 TroFuse-005 trial evaluated Sacituzumab Tirumotecan in 776 patients with advanced or recurrent endometrial cancer or carcinosarcoma whose disease had progressed after platinum-based chemotherapy and anti-PD-1/PD-L1 immunotherapy. The study compared the drug with physician’s choice chemotherapy, including doxorubicin or paclitaxel.
Researchers reported that Sacituzumab Tirumotecan significantly improved both overall survival and progression-free survival compared with standard chemotherapy. The trial also met its key secondary endpoint of objective response rate, demonstrating clinically meaningful antitumor activity in this difficult-to-treat patient population.
The treatment’s safety profile was consistent with previous studies, with no new safety concerns identified. Sacituzumab tirumotecan targets the TROP2 protein, which is commonly overexpressed in solid tumors, and delivers a topoisomerase I inhibitor payload directly to cancer cells using a specialized linker technology designed to reduce systemic toxicity.
