After Prior Treatments
A phase 1/2 study (2870-001/KL264-01) reports that sacituzumab tirumotecan (sac-TMT), a TROP2-targeting antibody-drug conjugate, shows strong antitumor activity in patients with advanced or metastatic urothelial carcinoma who have already failed standard treatments.
Among 49 heavily pretreated patients, the overall response rate was 31%, with higher responses (50%) in those who had only one prior line of therapy. The disease control rate reached 71%. Median progression-free survival was 5.5 months, and median overall survival was 12.1 months. Notably, the duration of response was not yet reached, suggesting lasting benefits in responders. The 18-month survival rate was 33%.
All patients experienced side effects, with 73% having severe reactions such as anemia, low neutrophil counts, and stomatitis. However, only 10% discontinued treatment. Overall, sac-TMT shows promise as a later-line option with manageable safety.
