IL-12 Immunotherapy Plus Liver-Targeted Chemotherapy Shows Strong Early Results in Metastatic Colorectal Cancer

A Phase 2 study shows that combining PDS01ADC, an IL-12 immunotherapy agent, with hepatic artery infusion pump (HAIP) chemotherapy may significantly improve outcomes for patients with metastatic colorectal cancer that has spread to the liver.

The combination aims to both directly attack liver tumors and stimulate the immune system. PDS01ADC helps activate immune responses against cancer, while HAIP delivers chemotherapy directly into the liver to intensify local tumor control.

In a small group of patients, the combination achieved a 77.8% response rate, compared with about 35% for HAIP chemotherapy alone. Two-year survival was also higher, at about 85% versus 40% in historical controls. Progression-free survival has not yet been reached after more than a year of follow-up.

Side effects were common but manageable, and no major liver complications were seen early in treatment. The trial is ongoing and expanding to confirm these encouraging results.
