CellCentric has treated the first patient in the DOMMINO-1 trial, a Phase 2 study testing an all-oral combination of inobrodib, pomalidomide, and dexamethasone (InoPd) for patients with heavily pretreated, relapsed or refractory multiple myeloma. The trial plans to enroll 100 patients in the US and UK, focusing on those who are “triple-class refractory” and often have failed prior bispecific or BCMA-targeted therapies. The main goal is to measure overall response rates.
Inobrodib works by targeting proteins that help myeloma cells survive, such as MYC and IRF4. Earlier studies showed a 60–69% response rate at the optimized 20 mg dose, nearly twice the historical rates for similar patients. The treatment is well tolerated, with a safety profile comparable to standard pomalidomide-dexamethasone therapy, making it suitable even for frail patients.
Being fully oral, the InoPd regimen avoids hospital-based infusions required by other modern myeloma treatments like bispecific antibodies or CAR T therapy. This makes it easier for patients treated in community settings or living far from specialized centers to access care.
